The 50-year-old Johnson opened with consecutive 67s and played the front nine in 5-under 30 before adding two birdies on the back nine for a 13-under 197 total.
He has two victories in eight senior starts this season.
"It was for the taking today. It was pretty much point and shoot and stop," Johnson said. " Miguel Angel Jiménez, 62, shot a 62 to finish second. The defending champion shot his age for the first time.
"Every time you can shoot your age is great," Jiménez said. Cameron Percy, the third-round leader, shot a 70 and stands third at 7 under. Darren Clarke and Tag Ridings each shot 68 to tie for fourth at 6 under.
Stewart Cink shot a 68 to tie for sixth at 5 under. Bernhard Langer, 68, shot a 67 and is tied for 14th at 3 under. He has now shot his age or better 45 times. Johnson said he played 54 good holes and has 18 more to play.
"You never know what could happen tomorrow depending on the conditions, wind or lack thereof," he said. This is the final year the tournament will be held at Firestone Country Club, ending a relationship with the PGA Tour that dates to the Rubber City Open in 1954.
